[IterateLineThroughImage example](https://itk.org/ITKExamples/src/Core/Common/IterateLineThroughImage/Documentation.html) seems close to what you want.

There are many examples for reading DICOM, [ReadDICOMSeriesAndWrite3DImage](https://itk.org/ITKExamples/src/IO/GDCM/ReadDICOMSeriesAndWrite3DImage/Documentation.html) is one of them.

Look into [Polygon Spatial Object](https://itk.org/Doxygen/html/classitk_1_1PolygonSpatialObject.html), in particular `IsInside*` methods.
